Molecular mechanism of activation of PLC-beta by Gq

Abstract

A broad range of agonists for GPCRs activate phospholipase C-beta through Gq family G proteins to regulate a variety of physiological process including nerutransmission, sensory perception, cardiovascular regulation, and cell growth. We have determined the crystal structure of complex of Galphaq and PLC-b3 at 2.7 angstrom. The structure revealed the molecular mechanism of activation of PLC-beta by Galphaq for the first time. Biochemical analysis of Galpha1-PLCb interaction also suggests the presence of multiple structures of Galphaq-PLCb complex.
